EXAMPLE: Enzymatic Assay of Glucose
Enzymatic assay of glucose, monitored at 505nm, is carried out by reaction rate measurement
during a stop flow period lasting 20 seconds, while data are collected for a calibration curve. Using a single stream flow instrument, a series of standards containing 500, 1000, 1500, 2000 and 2500 ppm glucose were injected, yielding the response curves shown on the right.
Note that the travel time between injector and detector is minimized, by using a short reaction coil to carry out the reaction within the flow cell while pump is stopped.
This experiment designed by Richard Wu, serves as the laboratory exercise , supervised by Tom Leach for the course in Instrumental Analysis at the Department of Chemistry, University of Washington, Seattle WA.
